In order to back up Ardour (prior to a system update, for example), is it sufficient to copy ~/.ardour2 plus the session folders? (And, obviously, restore them to their appropriate locations afterward…)

For your data and config yes, assuming you always embedded and didn’t link files. Linking files could be different, but it should be handled correctly so long as you copy the sessions to a different filesystem, but I haven’t checked this in some time myself.
For the software itself I would just redownload it from here myself.
